Beyond the Blood Test: A Closer Look at the Science of ELISpot Assays
When you have a routine blood test, it gives a broad snapshot of your health. But when a researcher needs to understand a specific immune response at a cellular level, they turn to more advanced tools like the ELISpot assay. ELISpot, which stands for Enzyme-Linked Immunospot, is a highly sensitive laboratory technique that is a staple in immunology research. The process begins with isolating a patient’s immune cells and placing them into a specialized plate. If a T-cell is activated by a specific antigen (like a component of a virus), it will secrete signaling molecules called cytokines. The plate is coated with a capture antibody that traps these secreted cytokines. A second detection antibody is then added, which binds to the trapped cytokines and, with the addition of a substrate, creates a visible "spot" for each activated cell.
